Effect of Chitim on Propranolol Hydrochioride Release from Press-coated Tablets.

Because of cellulose-like properties and insolubility in aqueous system, chitin was investigated as press-coating material to modify the release of propranolol hydrochloride from coated tablets. Effect of coating level, compression force and pH of dissolution fluid on drug release was tested and compared to that of ethylcellulose and microcrystalline cellulose (Avicel PH102). An enhancement of compression force and amount of coating layer during manufacturing of tablet coated with chitin could prolong drug release. However, the drug release was increased after the compression force was higher than 3,000 lbs. The drug release from tablets coated with chitin was pH dependent owing to the inherent property of chitin. The higher incorporation of chitin in phytowax layer could enhance the drug release. Nonlinear regression analysis was used to compare the degree of dissolution profile fitting to different mathematical release equations (power law, first order, Higuchi’s, zero order and KGT2). Determination of curve fitting with power law expression revealed that most drug release was anomalous. From parameters indicating degree-of-fit, the most suitable equation was first order, therefore, the drug diffusion through these press-coated layer into dissolution fluids depended on inner activity
