In searching for potent radical scavenging agents, a series of phenolic amides (1-4) were designed and synthesized. Four new amides were tested for the inhibitory action against lipid peroxidation using thiobarbituric acid method. All of the amides inhibited lipid peroxidation more than their corresponding acids. Ferulic amide 1 was the most active with IC50 of 19.92 uM. Investigation of the neuroprotection activity in NG 108-15 cells against H2O2 induced oxidative stress was performed. All new amides were able to reduce the number of cell deaths. Ferulic amide 1 was the most potent, showing significant effect at 1 nM (37.41% survival). The results indicated that, as proposed, the radical scavenging ability of the new amides was more potent than the corresponding acids. © All right reserved
