| บทคัดย่อ: Triamcinolone acetonide เป็น corticosteroid ชนิดหนึ่งที่ใช้รักษาอาการอักเสบที่ผิวหนังที่นิยมใช้กันอย่างแพร่หลายในรูปของยาครีมหรือขี้ผึ้ง โครงการพิเศษนี้เป็นการพัฒนาหาสูตรตำรับของ Triamcinolone acetonide ชนิดโลชั่นหรือครีมที่มีความหนืดน้อย เพื่อให้สะดวกแก่ผู้ป่วยที่จำเป็นจะต้องใช้ยาทาบริเวณกว้าง หรือในบริเวณที่มีเส้นขน หรือเส้นผมปกคลุมอยู่มาก โดยศึกษาหาตำรับที่มีความคงตัวทางกายภาพที่ดี และสามารถปลดปล่อยตัวยาได้ในระดับสูง วิธีทำการศึกษาโดยหา lotion base ที่มีความคงตัวดีโดยไม่เกิดการแยกชั้นระหว่างชั้นน้ำ และชั้นน้ำมัน เมื่อตั้งทิ้งไว้ที่อุณหภูมิห้องเป็นเวลา 1 เดือน และหลังจากทำการ freeze and thaw test จำนวน 5 cycles รวมทั้งมีคุณสมบัติทางกายภาพอื่น ๆ ที่ดีได้แก่ สี ความเนียน ความหนืด และความเป็นกรดด่างที่เหมาะสม จากนั้นจึงนำ lotion base ที่ได้มาเตรียมเป็น Triamcinolone acetonide lotion 0.1% และทำการทดสอบการปลดปล่อยตัวยาผ่าน cellophane membrane โดยใช้ Franz diffusion cell และใช้ ethanol 95% เป็น solvent จากการศึกษาพบว่าตำรับ Triamcinolone acetonide lotion ในความเข้มข้น 0.1% ที่มีความคงตัวดีและการปลดปล่อยตัวยาที่ดีที่สุด 3 ตำรับ ได้แก่ 1) ตำรับที่มีส่วนประกอบของ Triamcinolone acetonide, lanolin, cetyl,alcohol, mineral oil, brij 35 และ carbopol 9412) ตำรับที่มีส่วนประกอบของ Triamcinolone acetonide, soft paraffin, lanolin, steric acid, sodium lauryl sulfate และ veegum 3) ตำรับที่มีส่วนประกอบของ Triamcinolone acetonide, isopropyl myristate, glyceryl monostearate และ carbopol 934 |
| abstract: Triamcinolone acetonide, one of the moderate group of corticosteroids, is widely used topically for the skin inflammation. Its topical preparation is generally thick or viscous cream. The aim of this study is to develope a preparation of triamcinolone acetonide in low viscosity cream or lotion which might be useful for applying on large area or hairy skin. Twelve lotion bases were prepared and kept for one month at room temperature. They were then observed for any changes in their physical properties which were color, smoothness, viscosity and pH. The freeze and thaw test was also performed for 5 cycles. The lotion bases of four formulations which were most stable were selected to prepare 0.1% Triamcinolone acetonide lotion. Finally, these lotions were tested for the release of the drug from the lotion base by using the Franz diffusion cell. The results from the study show that the 0.1% Triamcinolone acetonide lotions which had good physical stability and high rate of release were : 1. formulation of triamcinolone acetonide, lanolin, cetyl alcohol, mineral oil, brij 35 and carbopol 941 2. formulation of triamcinolone acetonide, soft paraffin, lanolin, stearic acid, sodium lauryl sulfate and veegum 3. formulation of triamcinolone acetonide, isopropyl myristate, glyceryl monostearate and carbopol 934 |
