| บทคัดย่อ: โครงการพิเศษนี้จัดทำขึ้นเพื่อจำแนกสมุนไพรที่ใช้วิธีรับประทาน ในบัญชียาหลักแห่งชาติของไทยตามระบบการจำแนกตามชีวเภสัชกรรม (BCS ; Biopharmaceutics Classification System) โดยใช้โปรแกรม ADMET predictorTM ซึ่งการจำแนก BCS class เป็นวิธีการจำแนกสารที่ขึ้นอยู่กับค่าการซึมผ่าน และการละลายในระบบทางเดินอาหาร โดยสมุนไพรที่นำมาใช้จำแนกเป็นสมุนไพรที่ในบัญชียาหลักแห่งชาติ ตามบัญชียาจากสมุนไพร พ.ศ.2554 ในหัวข้อยาพัฒนาจากสมุนไพรและสมุนไพรที่คัดเลือกต้องเป็นสมุนไพรเดี่ยวที่ใช้วิธีรับประทานเท่านั้นซึ่งสมุนไพรที่ได้คัดเลือกมามีทั้งหมด 14 ชนิด ประกอบด้วย กล้วย, มะระขี้นก, บัวบก, ฟ้าทะลายโจร, หญ้าดอกขาว, ขิง, หญ้าหนวดแมว, เถาวัลย์เปรียง, รางจืด, ชุมเห็ดเทศ, กระเจี๊ยบแดง, มะขามแขก, ขมิ้นชัน และ หญ้าปักกิ่ง สำหรับการจำแนก BCS class โดยโปรแกรม ADMET predictorTM ต้องใช้โครงสร้างสารสำคัญในพืชแต่ละชนิด และปริมาณการใช้สูงสุดของสารสำคัญแต่ละตัว ในการวิเคราะห์ประเมินค่าที่ต้องการ จากนั้นมาวิเคราะห์ประเมินผ่านโปรแกรม ADMET predictorTM จนได้ค่าการซึมผ่าน และค่าการละลาย ในสภาวะ pH 3 ค่า คือ 1.2, 4.5 และ 6.8 จากนั้นนำค่าการละลายมาหา Dose number โดยใช้ปริมาณ volume คือ 250 ml และนำมาจำแนกเป็น class ต่างๆ ได้ตัวอย่างสารสำคัญที่อยู่ BCS class 1 จำนวน 17 ตัวอย่าง BCS class 2 จำนวน 10 ตัวอย่าง BCS class 3 จำนวน 65 ตัวอย่าง BCS class 4 จำนวน 12 ตัวอย่าง และไม่สามารถจำแนกได้ว่าเป็น class ใด class หนึ่งได้ จำนวน 5 ตัวอย่าง ซึ่ง ผลการจำแนก BCS class มีความสำคัญต่อการพัฒนาตำรับยาจากสมุนไพรต่อไปในอนาคตได้ |
| abstract: The aim of special project is classify of medicinal herbs taken orally listed in Thai National of Essential medicines using ADMET predictorTM software. Biopharmaceutics Classification system (BCS) are categorized into four biopharmaceutical classes according to both water solubility and membrane permeability. The total of fourteen herbs were chosen from the topic of medicines developed from medicinal herbs being listed in the List of Herbal medicinal Products in Thai National List of Essential medicines A.D 2011 and those selected medicinal herbs are taken orally without combination of the others. The medicinal herbs include Banana, Bitter gourd, Centella, Faa Tha Laal Jone, Feather grass, Ginger, Java tea, Jewel vine, Rangchuet, Ringworm bush, Roselle, Senna, Turmeric and Ya Pak King. ADMET predictorTM can predict permeability and water solubility values in 3 pH including pH 1.2, pH 4.5 and pH 6.8 in water 250 ml to obtain the dose numbers. Among the compounds predicted, there are 17 in BCS Class I, 10 in BCS Class II, 65 in BCS Class III, 12 in BCS Class IV and 5 in nonspecific BCS Class. This BCS classification of medicinal herbs are essential for formulation development in the future. |
